Learn MoreHuman primordial germ cells (PGCs) are precursors of sperm and oocytes and are crucial for the maintenance of the species. We found that the female PGCs have four sequential yet distinct developmental phases- mitotic, retinol acid signaling responsive, meiotic prophase, and oogenesis phase. Similarly we also identified three well-defined subpopulations for male PGCs. More importantly, one individual embryo usually contains several of these subpopulations simultaneously, highlighting the innate and highly entangled heterogeneity of PGC development in vivo. Moreover, we found the reciprocal signaling interactions between PGCs and their gonadal niche cells including KITL(soma)-KIT(PGC) and DLL(PGC)-NOTCH(soma) signaling pathways, as well as the signaling differences between female and male PGCs, which counterparts their sexual differentiation and the mitotic versus meiotic differences. Our works gives new insights of the crucial features of human early germ cells during their highly ordered mitotic, meiotic, and gametogenesis processes in vivo.
