Is the disease biology characterized well enough to anchor a program?

Disease biology at the molecular, cellular and genetic level — for example, pathway and biological program activation.

Target & Therapeutic Actionability

Is the target a tractable point of intervention in that biology?

Whether the target is present, accessible and specific in disease tissue — for example, target RNA expression.

Model & Disease Alignment

Full standard

Does the preclinical model recapitulate the patient biology the program depends on?

How closely the model reproduces the patient's disease biology — for example, immune context concordance.

Mechanistic & Functional Confidence

12 criteria

Does perturbing the target behave the way the hypothesis predicts?

Whether perturbing the target produces the predicted effect — for example, multi-omic biomarker consistency.
