GSE135690: Impact of sevoflurane priming on B16F10 transcriptome after in vivo growth

Bulk RNA sequencing

Murine skin melanoma cell line B16-F10 (ATCC no. CRL-6475) was obtained from the national standard repository (LGC Standards, Wesel, Germany). Priming of B16-F10 cells with sevoflurane was accomplished within a closed hypoxia chamber located in an 37C heating cabinet. Cell flasks were put into the chamber and the chamber was flushed with 2% sevoflurane (Baxter, Unterschleissheim, Germany) in normal air with 21% O2 for 30min, followed by an incubation for additional 2h. Subsequently, cells were prepared and injected into adult male C57BL/6 mice within 2h. Processing of the cells before injection was as described above. After a maximum of 4 weeks after injection, animals were euthanized and the tumor was resected for further analysis.
